EFFECT OF PHOTOBIOMODULATION AND STEM CELLS APPLICATION ON THE EXPRESSION OF MATRIX METALLOPROTEINASE (MMP) AND ITS INHIBITOR DURING ORTHODONTIC RELAPSE IN RATS

Abstract
Background: Relapse after orthodontic tooth development (OTM) happens through the rebuilding of the trans-shaped periodontal ligament (PDL), this rebuilding process involve the extracellular matrix (ECM), MMPs and tissue inhibitor metalloproteinases (TIMPs) may play a central role in regulating the production of ECM molecules. Some authors found that MMP2 and TIMP1 levels were transiently increased during orthodontic force application, the effect of low level laser therapy (LLLT) andor mesenchymal stem cells (MSCs) on the levels of MMP2 and TIMP1 was not determined. Materials and methods: The animals will be randomly divided into six groups each group consist of 10 rats :Group (A): without any intervention as a baseline control group Group (B): LLLT group, in which the low level laser was used to irradiate the mesial aspect of the root of maxillary central incisors. Group (C): MSCs group, in which (50000) MSCs were applied on the mesial aspect of the root of maxillary central incisors. Group (D): MSCs, in which (100000) MSCs were applied on the mesial aspect of the root of maxillary central incisors. Group (E): MSCs and LLLT group, in which (50000) MSCs were applied on the mesial aspect of the root of maxillary central incisors and will be irradiated by LLLT. Group (F): MSCs and LLLT group, in which (100000) MSCs were applied on the mesial aspect of the root of maxillary central incisors and will be irradiated by LLLT. The concentrations of MMP2 and TIMP1 in ratís blood serum were detected using Enzyme-linked immunosorbent assay (ELISA). Results: the results showed that the LLLT and MSCs groups showed the highest expression values for TIMP1. Whereas, the MSCs groups revealed elevated expression values for MMP2. Conclusion: LLLT stimulate the expression of TIMP1 which enhance PDL remodeling and could decrease the orthodontic relapse. MSCs showed their repair ability during the 2 weeks period by increased MMP2TIMP1 ratio.
